目的 研究阿米洛利 (amiloride ,Amil)对甲状腺素诱发大鼠肥厚心肌钙电流的作用。方法 大鼠ipL 甲状腺素造成心肌肥厚模型后 ,用膜片钳技术记录L型钙电流。结果 肥厚组心肌细胞钙电流幅度、细胞膜电容增加 ,半数激活电压 (V1/ 2 )向负电位方向移动 ,斜率 (k)减小。Amil治疗后使肥厚细胞钙电流幅度、细胞膜电容及半数激活电压的改变均朝正常方向变化。预先给予Amil后 ,血管紧张素II和苯肾上腺素不表现出促钙作用。结论Amil能对抗甲状腺素致肥厚的心肌细胞钙电流幅度与膜电容的增大 ,还可对抗血管紧张素II和苯肾上腺素对钙电流的增加以发挥拮抗致肥厚因子的作用。
Objective To investigate the effect of amiloride (Amil) on thyroxin-induced calcium currents in hypertrophic myocardium of rats. Methods After rat model of myocardial hypertrophy induced by ipL thyroxine, L type calcium current was recorded by patch clamp technique. Results In the hypertrophic group, the amplitude of calcium current and membrane capacitance of cardiomyocytes increased. The half activation voltage (V1 / 2) shifted toward the negative potential and the slope (k) decreased. After Amil treatment, the change of calcium current amplitude, cell membrane capacitance and half activation voltage of hypertrophic cells all changed in the normal direction. After Amil administration, angiotensin II and phenylephrine did not show a calcium-stimulating effect. Conclusion Amil can counteract the increase of calcium current amplitude and membrane capacitance induced by thyroxine in hypertrophic cardiomyocytes, and antagonize the increase of calcium current induced by angiotensin II and phenylephrine to antagonize hypertrophic factors.
