目的:观察心脑宁片对D-半乳糖致小鼠亚急性衰老模型胸腺组织形态的影响。方法:取体质量18~21 g小鼠72只,雌雄各半,其中12只为空白对照组,余60只每天颈背部皮下注射D-半乳糖(生理盐水配成质量浓度为5%,注射量为0.025m L·g-1)制作糖代谢衰老模型,每天给药1次,连续40 d。模型制作成功后,随机平均分为模型组(生理盐水0.02 m L·g-1),脑安片组(0.53 g·kg-1,26.5 g·L-1,0.02 m L·g-1,相当于临床用量的15倍),大剂量、中剂量、小剂量心脑宁片组(2.24 g·kg-1、1.12 g·kg-1、0.56 g·kg-1,112 g·L-1、56 g·L-1、28 g·L-1,0.02 m L·g-1,相当于临床剂量的30倍、15倍、7.5倍)。每天给药1次,连续30 d。取小鼠胸腺组织用福尔马林固定,石蜡包埋,切片,HE染色,光镜观察胸腺组织形态的变化。结果:模型组胸腺皮质厚度、淋巴细胞数分别为(7.26±2.18)μm、(21.4±3.7)个,脑安片组分别为(23.18±9.45)μm、(36.3±12.2)个,大剂量心脑宁片组分别为(27.89±3.46)μm、(46.6±10.4)个,中剂量心脑宁片组分别为(24.36±4.19)μm、(37.5±8.60)个,小剂量心脑宁片组分别为(22.18±3.45)μm、(36.3±7.7)个,空白对照组分别为(28.62±4.35)μm、(57.3±11.4)个。各组胸腺皮质厚度、淋巴细胞数与模型组比较,差异均有统计学意义(P<0.01)。结论:心脑宁片可显著增加D-半乳糖致小鼠亚急性衰老模型胸腺组织的厚度和淋巴细胞数,有较好的抗衰老的作用。
Objective: To observe the effects of Xinnaoning Tablet on the morphology of thymus in D-galactose induced subacute aging mice. Methods: Seventy-two mice with 18 ~ 21 g body mass were divided into two groups, male and female, of which 12 were blank control group. The remaining 60 rabbits were injected subcutaneously with D-galactose (5% The amount of 0.025m L · g-1) made glucose metabolism aging model, once a day for 40 days. Administered once daily for 30 days. The thymus tissue of mice was fixed with formalin, embedded in paraffin, sectioned and stained with HE. The morphology of thymus was observed with light microscope. Results: The thickness of thymic cortex and the number of lymphocytes in the model group were (7.26 ± 2.18) μm and (21.4 ± 3.7) respectively, respectively, while that in the Naogan Tablet group was (23.18 ± 9.45) μm and (36.3 ± 12.2) (27.89 ± 3.46) μm and (46.6 ± 10.4), respectively, in the medium-dose Xinnaoning tablet group were (24.36 ± 4.19) μm and (37.5 ± 8.60) (22.18 ± 3.45) μm and (36.3 ± 7.7) mm respectively. The blank control group was (28.62 ± 4.35) μm and (57.3 ± 11.4) mm respectively. The thymus cortex thickness and the number of lymphocytes in each group compared with the model group, the difference was statistically significant (P <0.01). Conclusion: Xinnaoning Tablet can significantly increase the D-galactose-induced hypothyroidism in mice thymus tissue thickness and lymphocyte number, have a good anti-aging effect.
