目的研究不同剂量亚砷酸钠慢性饮水染毒大鼠肝脏纤维化情况。方法将40只健康8周龄SPF级SD雄性大鼠随机分为4组,分别为对照(去离子水)组及0.68、1.36、2.73mg/kg亚砷酸钠染毒组。采用自由饮水方式进行染毒,连续染毒24周。测定血清肝功能指标丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和肝纤维化指标[血清中透明质酸(HA),Ⅲ型前胶原(PCⅢ),Ⅳ型胶原(Ⅳ-C),层粘连蛋白(LN)],并观察病理学变化。结果亚砷酸钠染毒20周时2.73 mg/kg组大鼠体重小于对照组(P<0.05),24周时2.73 mg/kg组大鼠体重小于对照组、0.68、1.36 mg/kg组(P<0.05)。0.68 mg/kg组未见明显的形态学改变,1.36、2.73 mg/kg组随染毒剂量升高肝纤维化病变明显。1.36、2.73 mg/kg组AST活力均高于对照组和0.68 mg/kg组(P<0.05)。各染毒组ALT活力高于对照组,2.73 mg/kg组高于0.68、1.36 mg/kg组(P<0.05)。HA、PCⅢ、LN三项指标1.36、2.73 mg/kg组均高于对照组和0.68 mg/kg组,且2.73 mg/kg组高于1.36 mg/kg组,差异有统计学意义(P<0.05)。1.36、2.73 mg/kg组Ⅳ-C高于对照组和0.68 mg/kg组(P<0.05)。结论慢性砷暴露后砷在肝脏有蓄积,导致肝脏纤维化且肝功能下降。
Objective To study the hepatic fibrosis in rats exposed to sodium arsenite at different dosages. Methods Forty SD male SD rats of 8 weeks old were randomly divided into 4 groups: control (deionized water) group and 0.68, 1.36, 2.73 mg / kg sodium arsenite exposure group. Using free drinking water for exposure, continuous exposure to 24 weeks. Serum liver function indicators such as alanine aminotransferase (ALT), aspartate aminotransferase (AST) and indicators of liver fibrosis [serum hyaluronic acid (HA), type III procollagen (PCIII) Collagen (Ⅳ-C) and laminin (LN)] were observed and pathological changes were observed. Results At 20 weeks after arsenite treatment, the body weight of 2.73 mg / kg group was less than that of the control group (P <0.05). At 24 weeks, the body weight of 2.73 mg / kg group was less than that of the control group, 0.68 and 1.36 mg / kg group P <0.05). 0.68 mg / kg group no obvious morphological changes, 1.36,2.73 mg / kg group with the dose increased liver fibrosis lesions significantly. The activities of AST in 1.36,2.73 mg / kg group were higher than those in control group and 0.68 mg / kg group (P <0.05). The activity of ALT in each exposure group was higher than that in the control group, but higher in the 2.73 mg / kg group than in the 0.68 and 1.36 mg / kg groups (P <0.05). The indexes of HA, PCⅢ and LN in 1.36,2.73 mg / kg group were higher than those in control group and 0.68 mg / kg group, and those in 2.73 mg / kg group were higher than those in 1.36 mg / kg group (P <0.05 ). The levels of Ⅳ-C in 1.36,2.73 mg / kg group were higher than those in control group and 0.68 mg / kg group (P <0.05). Conclusion Arsenic accumulated in the liver after chronic arsenic exposure, resulting in hepatic fibrosis and decreased liver function.
