目的 为寻找有效的抗日本血吸虫感染疫苗候选抗原分子。方法 用紫外线照射致弱尾蚴免疫兔血清筛选日本血吸虫成虫cDNA文库 ,再用同法免疫鼠脾细胞制备的单克隆抗体对所获阳性克隆进行再筛选 ,并对免疫筛选的阳性克隆的cDNA插入片段进行PCR扩增。结果 兔免疫血清确定 12个阳性克隆 ,其中 6个克隆与 2株以上的单抗呈阳性反应 :PCR扩增cDNA插入片段大小约 40 0bp~ 1 4kb左右。结论 已筛选到与致弱尾蚴免疫兔血清及同法免疫制备单抗产生特异反应的阳性克隆 ,获得一批编码可能具有保护性作用的日本血吸虫抗原的基因片段。
The purpose is to find effective anti-Japanese Schistosoma vaccine candidate antigen molecules. Methods The cDNA library of adult Schistosoma japonicum cDNA library was screened by immunizing rabbit with weak cercariae irradiated with ultraviolet light. The positive clones were screened by the monoclonal antibody prepared from the splenocytes of mice immunized with the same method. The positive clones PCR amplification. Results Twelve positive clones were identified in rabbit serum. Six of the clones showed positive reaction with two or more McAbs. The size of cDNA insert was about 40 0bp ~ 14kb. Conclusion A number of positive clones that specifically react with immunized rabbits immunized with weak cercariae and monoclonal antibodies produced by the same method were screened. A number of gene fragments encoding for the possible protective effect of Schistosoma japonicum antigen were obtained.
