为了满足日益增长的冬油菜新品种鉴定需要,采用多色SSR荧光标记组合毛细管电泳检测法,构建了419份2013-2015年度国家冬油菜区域试验中的冬油菜新品种(系)×38对SSR荧光标记的DNA指纹数据库。所有标记共检测到41个位点,其中双位点标记3个,单位点标记35个。38个多态性SSR标记于419份冬油菜新品种中共检测到129个等位变异;每个位点的变异范围为2~6,平均每个位点3.15个。等位基因型数共250个,每个位点的基因型数为3~15个,平均每个位点6.09个。41个位点的多态性信息指数PIC值变幅为0.132 2~0.690 7,平均为0.390 2。统计指纹数据库缺失率为1.1%,具有较好的完整性。DICE相似系数矩阵分析得出,419份中国冬油菜新品种(系)间成对遗传相似系数共获取87 571个数据项,品种间遗传相似系数变幅为0.366~0.983,平均遗传相似系数为0.692。相似系数在区间0.601~0.800内共有78 571个数据项,占总数的89.72%。此结果一定程度表明冬油菜新品种(系)的遗传多样性不够丰富。
In order to meet the need of identifying new winter rapeseed varieties, 417 new winter rape varieties (lines) in the National Winter Rapeseed Regional Trial 2013 ~ 2015 were constructed by using the multi-color SSR fluorescent marker combination capillary electrophoresis assay. Fluorescently labeled DNA fingerprinting database. A total of 41 loci were detected in all markers, including 3 double loci and 35 single loci. Thirty-eight polymorphic SSR markers were detected in 419 new winter rape cultivars. The variation range of each locus ranged from 2 to 6 with an average of 3.15 per locus. The total number of alleles was 250, and the number of genotypes per locus ranged from 3 to 15 with an average of 6.09 loci per locus. The polymorphism information index of 41 loci PIC ranged from 0.132 2 to 0.690 7 with an average of 0.390 2. Statistical fingerprint database deletion rate of 1.1%, with good integrity. DICE similarity coefficient matrix analysis showed that 87 571 pairs of genetic similarity coefficients were obtained among 419 Chinese winter rape cultivars (lines). The genetic similarity coefficients of cultivars ranged from 0.366 to 0.983 with an average genetic similarity coefficient of 0.692 . The similarity coefficient has 78 571 items in the range of 0.601 ~ 0.800, accounting for 89.72% of the total. To some extent, this result indicated that the genetic diversity of new winter rape varieties (lines) was not rich enough.
