目的研究超声波与丙醇-硫酸铵双水相耦合从3种提取分离黄酮的工艺条件及参数及提取物抗氧化活性。方法以Al(NO_3)_3-NaNO_2分光光度法测定总黄酮,研究丙醇-硫酸铵双水相形成条件及超声提取条件对总黄酮提取率的影响,优化提取条件,以Fe~(2+)为脂质过氧化促进剂,以卵磷脂脂质体为模型研究提取物抗氧化活性。结果对3种中药中总黄酮的提取率在2.7%~6.12%之间,与回流提取法相近,而提取物中总黄酮含量为17.2%~21.1%,明显高于回流提取法,提取物对脂质过氧化最大抑制率在70.1%~74.4%之间。结论方法可有效地从中药中提取黄酮,提取物对脂质过氧化具有良好的抑制作用,方法操作简便、条件温和,环境友好,适于中药中黄酮的提取。
Objective To study the extraction conditions and parameters of extraction and separation of flavonoids from ultrasonic coupling with propanol-ammonium sulphate aqueous solution and their antioxidant activity. Methods Al(NO_3)_3-NaNO_2 spectrophotometric method was used to determine total flavonoids. The effects of the formation conditions of the aqueous two-phase propanol-ammonium sulfate and ultrasonic extraction conditions on the extraction rate of total flavonoids were studied. The extraction conditions were optimized to Fe 2+ As a lipid peroxidation enhancer, the antioxidant activity of the extract was studied using a lecithin liposome model. Results The extraction rates of total flavonoids in the three Chinese herbs were between 2.7% and 6.12%, similar to the reflux extraction method, and the content of total flavonoids in the extract was 17.2% to 21.1%, which was significantly higher than that of the reflux extraction method. The maximum inhibition rate of lipid peroxidation was between 70.1% and 74.4%. Conclusion The method can effectively extract flavonoids from traditional Chinese medicines. The extracts have a good inhibitory effect on lipid peroxidation. The method is simple, mild and environmentally friendly. It is suitable for the extraction of flavonoids from traditional Chinese medicine.
