目的探讨Wnt5a在早期雌性生殖管道发育过程中的表达。方法正常孕15.5d、17.5d、19.5d和21.5d的小鼠胚胎,PCR鉴定性别,取雌性胎鼠石蜡包埋,连续横断切片,每4张行1张HE染色,1张白片待用。HE染色后获取连续切片图像,根据雌性胚鼠生殖管道解剖走形,识别中肾旁管/子宫解剖定位。选取含有中段中肾旁管/子宫的白片,用SABC免疫组织化学方法,检测Wnt5a在不同发育时期雌性生殖管道的表达。结果获取含雌性生殖管道的鼠胚切片。同时检测到Wnt5a在孕15.5d到21.5d的雌性鼠胚中段中肾旁管/子宫上皮细胞、间质细胞均有表达,随着鼠胚发育,Wnt5a在间质细胞表达不断增强,差异在统计学上有极显著意义(P<0.01)。结论Wnt5a在雌性小鼠生殖管道形成过程中发挥作用,可能是诱导子宫发育及分化的关键因子。
Objective To investigate the expression of Wnt5a in early female reproductive tract development. Methods The normal pregnant 15.5d, 17.5d, 19.5d and 21.5d mouse embryos were identified by PCR. Female fetuses were embedded in paraffin and transected serially. HE staining was performed on every 4 rows and 1 white tablet was used . Serial sections were obtained after HE staining. According to the anatomical shape of the reproductive duct of female embryos, anatomical location of the para-renal tubular / uterus was identified. Select the middle of the paraventricular / uterine white tablets, using SABC immunohistochemical methods to detect Wnt5a in different developmental stages of female reproductive tract expression. Results Obtain embryo slices containing female reproductive duct. Wnt5a was also detected in the paraventricular nucleus / uterine epithelial cells and interstitial cells in the middle part of female rat embryos between 15.5d and 21.5d, and the expression of Wnt5a in stromal cells increased with the development of mouse embryos There is significant significance in learning (P <0.01). Conclusion Wnt5a plays a role in the reproductive tract formation of female mice and may be the key factor to induce uterine development and differentiation.
