一种抗TAG-72单克隆抗体CC49的单链抗原结合蛋白片段sFvCC49已经研究开发出来,并且在动物实验中显示对结直肠癌肝转移具有良好前景。 研究了5例结直肠癌肝转移病人手术前的情况。高效液相色谱分析显示低水平的积聚(结合物低于10%)。放射标记示踪前后,用干燥~(123)Ⅰ加磷酸盐缓冲剂,用稀盐酸滴定至pH值7.0。病人分别注射762MBq、444MBq、999MBq、925MBq和37MBq,并标记5mg的片段。用单光子发射CT于注射后4~6小时和24小时进行全身扫描。
A single-chain antigen binding protein fragment sFvCC49 against the TAG-72 monoclonal antibody CC49 has been studied and developed, and has shown promising prospects for liver metastasis of colorectal cancer in animal experiments. Five preoperative cases of colorectal liver metastases were studied. HPLC analysis showed low levels of accumulation (conjugates below 10%). Before and after radiolabeling, dry ~123I plus phosphate buffer and titrate with dilute hydrochloric acid to pH 7.0. Patients were injected with 762 MBq, 444 MBq, 999 MBq, 925 MBq, and 37 MBq, respectively, and 5 mg fragments were labeled. Whole body scans were performed using single-photon emission CT at 4 to 6 hours and 24 hours after injection.
