目的:观察益气活血通络方对db/db小鼠胰腺组织c-Jun氨基末端激酶(JNK)信号通路及炎症因子表达的影响,探讨益气活血通络方对胰岛细胞炎性损伤的保护作用及其机制。方法:用自发性2型糖尿病C57BL/Ks J-db/db小鼠为动物模型,将db/db小鼠随机分成模型组、硫辛酸组(0.078g/kg)、益气活血通络方6.4、3.2、1.6g/kg剂量组,另取10只db/m小鼠为正常对照组,连续灌胃给药8周,模型组给予生理盐水。药前、药后4周和8周测定小鼠血糖。实验结束后,摘取胰腺组织,HE染色法观察病理变化,ELISA法检测小鼠胰腺组织胰岛素含量,Western bloting与免疫组织化学法检测IL-1β、TNF-α、JNK、p-JNK蛋白的表达。结果:与正常组比较,模型组胰岛细胞出现严重的病理性损伤,药后4周和8周血糖、胰腺组织中IL-1β、TNF-α、P-JNK蛋白的表达水平明显高于正常组;与模型组比较,益气活血通络方组胰岛细胞病理损伤程度减轻,血糖降低,胰腺组织胰岛素含量升高,胰腺组织IL-1β、TNF-α、P-JNK蛋白表达水平显著下降。结论:益气活血通络方能够减轻db/db小鼠胰岛细胞损伤,可能与降低炎症因子表达,抑制JNK信号通路过度激活有关。
AIM: To observe the effect of Yiqi Huoxue Tongluo Prescription on c-Jun N-terminal kinase (JNK) signaling pathway and inflammatory cytokines expression in pancreatic tissues of db / db mice and to explore the protective effects of Yiqi Huoxue Tongluo Recipe against pancreatic islet inflammatory injury Role and mechanism. Methods: The db / db mice were randomly divided into model group, lipoic acid group (0.078g / kg), Yiqi Huoxue Tongluo Formula 6.4 , 3.2,1.6 g / kg dose group, another 10 db / m mice as normal control group, continuous gavage for 8 weeks, the model group was given normal saline. Pre-drug, drug 4 weeks and 8 weeks after the determination of blood glucose. After the experiment, the pancreatic tissues were harvested, the pathological changes were observed by HE staining, the contents of insulin in the pancreas tissue were detected by ELISA, the expressions of IL-1β, TNF-α, JNK and p-JNK protein were detected by Western blotting and immunohistochemistry . Results: Compared with the normal group, the islet cells in the model group showed severe pathological injury. The levels of IL-1β, TNF-α and P-JNK in the blood glucose and pancreatic tissues were significantly higher than those in the normal group at 4 weeks and 8 weeks Compared with model group, the pathological damage of islet cells in Yiqi Huoxue Tongluo Recipe group was alleviated, the blood glucose was decreased and the content of insulin in pancreas was increased. The expression of IL-1β, TNF-α and P-JNK in pancreatic tissue was significantly decreased. Conclusion: Yiqi Huoxue Tongluo Recipe can reduce islet cell injury in db / db mice, which may be related to the decrease of inflammatory factor expression and the inhibition of JNK signaling pathway over-activation.
