目的探讨E-钙黏蛋白及ZEB1在宫颈鳞癌及宫颈上皮内瘤变(CIN)组织中的表达及临床意义,了解二者在宫颈癌发生、发展及浸润转移过程中的作用。方法采用免疫组织化学SP法检测E-钙黏蛋白和ZEB1在59例宫颈鳞癌、43例CIN和30例正常宫颈组织中的表达情况,分析二者与宫颈鳞癌组织临床病理特征的关系。结果 E-钙黏蛋白在正常宫颈组织中的表达水平高于CINⅡ-Ⅲ级组织和宫颈鳞癌组织(P<0.05)。ZEB1在宫颈鳞癌组织、CINⅡ-Ⅲ级组织及CINⅠ级组织中的阳性表达率高于正常宫颈组织(P<0.05)。E-cadherin及ZEB1的表达强度与临床分期、淋巴转移、组织学分级等因素有相关性(P<0.05),与患者年龄无相关性(P>0.05)。E-钙黏蛋白在宫颈鳞癌组织中的异常表达与ZEB1的阳性表达呈负相关(r=-0.481,P<0.05)。结论 E-钙黏蛋白的表达缺失或降低与ZEB1过表达均在宫颈鳞癌的发生、发展及淋巴结转移过程中发挥重要作用。
Objective To investigate the expression and clinical significance of E-cadherin and ZEB1 in cervical squamous cell carcinoma and cervical intraepithelial neoplasia (CIN), and to understand the role of E-cadherin and ZEB1 in carcinogenesis, development and invasion and metastasis of cervical cancer. Methods The expressions of E-cadherin and ZEB1 in 59 cases of cervical squamous cell carcinoma, 43 cases of CIN and 30 cases of normal cervical tissue were detected by immunohistochemical SP method. The relationship between E-cadherin and ZEB1 in clinicopathological features was analyzed. Results The expression of E-cadherin in normal cervical tissue was higher than that in CINⅡ-Ⅲ and cervical squamous cell carcinoma (P <0.05). The positive rate of ZEB1 in cervical squamous cell carcinoma, CINⅡ-Ⅲstage and CINⅠstage was higher than that in normal cervical tissue (P <0.05). The expression of E-cadherin and ZEB1 was correlated with clinical stage, lymph node metastasis, histological grade and other factors (P <0.05), but not with patient’s age (P> 0.05). The abnormal expression of E-cadherin in cervical squamous cell carcinoma was negatively correlated with the positive expression of ZEB1 (r = -0.481, P <0.05). Conclusion The loss or decrease of E-cadherin expression and the overexpression of ZEB1 play an important role in the carcinogenesis, development and lymph node metastasis of cervical squamous cell carcinoma.
