目的 :检测Fas基因表达并观察转染Fas基因对卵巢癌细胞凋亡的影响 ,探讨其在卵巢癌发生、发展中的作用。方法 :经流式细胞术、RT PCR方法检测 6种卵巢癌细胞Fas的表达水平。构建pcDNA3 Fas真核表达载体 ,经脂质体转染细胞 ,通过流式细胞仪检测Fas基因表达变化。应用PI染色经流式细胞术检测观察转染Fas基因对3AO细胞凋亡的影响。结果 :6种卵巢癌细胞均表达Fas,均属中低表达。经 pcDNA3 Fas真核表达载体转染的卵巢癌细胞Fas基因表达及对Fas单抗诱导凋亡的敏感性均有不同程度提高。结论 :Fas基因低表达及对Fas介导凋亡的抵抗可能与卵巢癌发生、发展有关 ;转染Fas基因可提高Fas基因表达 ,可促进卵巢癌细胞凋亡
Objective: To detect the expression of Fas gene and observe the effect of Fas gene transfection on the apoptosis of ovarian cancer cells, and to explore its role in the occurrence and development of ovarian cancer. Methods: The expression of Fas in six kinds of ovarian cancer cells was detected by flow cytometry and RT-PCR. The eukaryotic expression vector pcDNA3 Fas was constructed and transfected into cells by lipofectamine. The expression of Fas gene was detected by flow cytometry. The effect of Fas gene transfection on 3AO cell apoptosis was observed by flow cytometry with PI staining. Results: All the six kinds of ovarian cancer cells expressed Fas, which were low expression. The expression of Fas gene in ovarian cancer cells transfected with pcDNA3 Fas eukaryotic expression vector and the sensitivity of Fas monoclonal antibody to apoptosis were all increased to some extent. Conclusion: The low expression of Fas gene and its resistance to Fas-mediated apoptosis may be related to the occurrence and development of ovarian cancer. The transfection of Fas gene may increase the expression of Fas gene and promote the apoptosis of ovarian cancer cells
