目的研究环六亚甲基双乙酰胺(HMBA)对胃癌细胞MGc80-3增殖分化的影响。方法使用3H-TDR标记DNA技术来显示HMBA诱导处理前后MGc80-3细胞内DNA合成量的变化。结果HMBA抑制MGc80-3细胞的生长活动,最高抑制率可达56.7%,细胞分裂指数降低1.25倍。细胞核内可标记的DNA量急剧下降,其细胞总DNA与对照组DNA的放射性CPM比值约为1∶5.22。结论HMBA通过抑制胃癌细胞MGc80-3的DNA合成进而抑制该细胞的增殖活动,从而达到诱导分化的效果。
Objective To study the effect of cyclohexamethylene bisacetamide (HMBA) on the proliferation and differentiation of gastric cancer cell line MGc80-3. Methods 3H-TDR labeled DNA technology was used to show the changes of DNA synthesis in MGc80-3 cells before and after HMBA induction. Results HMBA inhibited the growth of MGc80-3 cells with the highest inhibition rate of 56.7% and the cell division index decreased by 1.25 times. The amount of DNA that can be labeled in the nucleus drastically decreased. The ratio of the radioactive CPM of the total DNA of the cells to the DNA of the control group was approximately 1:5.22. Conclusion HMBA inhibits the proliferation of gastric cancer cells by inhibiting the DNA synthesis of MGc80-3, so as to achieve the effect of inducing differentiation.
