采用硫酸铵法、辛酸二步法、DEAE-Sephadex A50 离子交换柱层析法,从杂交瘤小鼠腹水中纯化人型结核杆菌H37Rv 株特异性抗原TB4 的单克隆抗体,从回收率、纯度和活性3 个方面进行比较。结果显示,硫酸铵法回收率最高,层析法纯度最高,3 种方法对单抗的活性均无大的影响。认为:辛酸二步法可有效地去除白蛋白,适用于大量腹水中单抗的提取、纯化,具有简单、省时等优点;柱层析法适用于小量腹水单抗纯化及分析用途
The monoclonal antibodies against Mycobacterium tuberculosis H37Rv strain TB4 were purified from ascites of hybridoma mice by ammonium sulfate method, two-step caprylic acid method and DEAE-Sephadex A50 ion exchange column chromatography. The recovery rate, purity and Activity of three aspects of comparison. The results showed that the highest recovery rate of ammonium sulfate, chromatography highest purity, the three methods have no significant impact on the activity of monoclonal antibodies. It is concluded that caprylic acid two-step method can effectively remove albumin and is suitable for the extraction and purification of monoclonal antibodies in a large amount of ascites, which has the advantages of simplicity, time saving, etc. The column chromatography is suitable for the purification and analysis of small amount of ascites
