目的研究川芎挥发油提取及其β-环糊精包合物制备的最佳工艺。方法采用L9(34)正交试验对影响川芎挥发油的提取工艺的提取时间、药材粒度、加水量等条件进行了优化实验;以投料比、包合温度、包合时间为考察因素,以包合物收率为考察指标,优选最佳包合工艺,通过薄层层析对包合物进行鉴定。结果确定最佳提取工艺条件为D2A2C3B3:即提取时间8 h,药材为过10目筛的最粗粉,浸泡2h,加水量为10倍。环糊精包合的最佳工艺为B2C3A2D1,即包合温度50℃,搅拌3 h,投料比为1:10,搅拌速度为低速。结论确定了川芎挥发油的最佳提取工艺,并优选出了其包合物的最佳包合工艺,经验证包合物与混合物有明显区别。
Objective To study the extraction of volatile oil from Ligusticum chuanxiong Hort and to optimize the preparation of β-cyclodextrin inclusion compound. Methods The orthogonal experiment L9 (34) was used to optimize the extraction conditions affecting the extraction process of volatile oil of Ligusticum chuanxiong Hort, the particle size of medicinal materials and the amount of water added. Taking feed ratio, inclusion temperature and inclusion time as the investigation factors, The recovery rate was the index of investigation, the best inclusion process was optimized, and the inclusion complex was identified by TLC. The results showed that the best extraction process conditions were D2A2C3B3: the extraction time was 8 h, the crude drug was over the 10 mesh sieve, soaked for 2 h, the amount of water was 10 times. Cyclodextrin inclusion process for the best B2C3A2D1, that inclusion temperature 50 ℃, stirring 3 h, the feed ratio of 1:10, the stirring speed is low speed. Conclusion The optimum extraction process of the volatile oil of Ligusticum chuanxiong Hort. Was determined, and the optimal inclusion process of its inclusion was optimized. The obvious difference was found between the inclusion compound and the mixture.
