目的 观察人类补体衰变加速因子 (DAF)在小鼠细胞表达后 ,对小鼠细胞抗人血清补体破坏的保护作用。方法 用人DAFcDNA转染小鼠细胞使该基因在小鼠细胞中表达。再将此种小鼠细胞放入含正常人血清的培养基中孵育 ,观察小鼠细胞是否被破坏。结果 转染人DAFcDNA并表达人DAF的小鼠细胞 ,在含人正常血清的培养基中孵育 48h后仍能存活并可以传代 ,而对照组的小鼠细胞孵育后在 1~ 4h内迅速死亡。结论 人类的DAF可以保护小鼠细胞不受补体的破坏。
Objective To observe the protective effect of human complement decay accelerating factor (DAF) on the anti-human serum complement destruction in mouse cells after its expression. Methods Transfection of mouse cells with human DAF cDNA allowed the gene to be expressed in mouse cells. The mouse cells were then incubated in medium containing normal human serum to observe whether the mouse cells were destroyed. Results The mouse cells transfected with human DAF cDNA and expressing human DAF survived and could be passaged 48 h after incubation with human normal serum, while the control mice died rapidly within 1 h to 4 h after incubation. Conclusion Human DAF protects mouse cells from complement damage.
