在前一篇(Shukla 和 Gough,1979)我们报道了检测二种线状植物病毒甘蔗花叶病毒(SCMV)和烟草花叶病毒(TMV)改良的流行免疫电镜技术(Derrick,1973;Milne 和 Luisoni,1977)。它包括在用特异抗血清包被铜网前,预先用 A 蛋白(一种金黄葡萄球菌细胞壁蛋白)包被铜网。虽然这条件对严格比较来说还不是最理想的,但此方法比起单用抗血清处理诱捕的 SCMV和 TMV 病毒粒体的数量多。此技术特别适合在植物抽提液中含病毒浓度低的病毒。
In the previous article (Shukla and Gough, 1979) we report the epidemiological immunoelectron microscopy technique for the detection of modified filamentous plant virus sugarcane mosaic virus (SCMV) and tobacco mosaic virus (TMV) (Derrick, 1973; Milne and Luisoni , 1977). It involves coating the copper mesh with protein A (a Staphylococcus aureus cell wall protein) in advance before coating the copper mesh with the specific antiserum. Although this condition is not ideal for a rigorous comparison, this method is more numerous than entrapped SCMV and TMV virions by antiserum alone. This technique is particularly suitable for containing low concentrations of virus in plant extracts.
