目的 2次回归旋转组合设计优化复合酶解法提取锁阳总糖工艺。方法以锁阳总糖得率为指标,酶解比例、酶用量、酶解温度、酶解时间、溶液初始p H值为考察因素,筛选出主要因素,并采用2次旋转组合设计方法优化提取工艺条件。采用响应面分析法(RSM),评价各因素水平的单因素效应、互作效应对响应值的影响。结果复合酶解法提取锁阳总糖最佳工艺:复合酶用量1%(锁阳粉末),酶解初始p H值5.5,酶解温度55℃,酶解时间45 min,果胶酶和纤维素酶比例1∶1,在此条件下锁阳总糖得率为(17.54±0.35)%。结论复合酶法提取锁阳总糖是一种简单有效的辅助方法,2次旋转正交组合设计法预测性良好。

Results The optimum conditions for extracting total sugars from Cynomorium acutissima were as follows: compound enzyme 1% (Cynomorium powder), initial p H value 5.5, enzymolysis temperature 55 ℃, hydrolysis time 45 min, pectinase and cellulose Enzyme ratio 1: 1, Under this condition, the total sugar yield was (17.54 ± 0.35)%. Conclusion The compound enzymatic extraction of total catalase is a simple and effective adjunct method. The 2-rotation orthogonal design method has good predictability.
