目的:研究三氧化二砷(As2O3)与维生素C联用对喉癌细胞(Hep-2)凋亡的协同作用。方法:As2O3及联用VitC孵育体外培养的人Hep-2,采用四甲基偶氮唑蓝(MTT)法测定细胞存活率,Annexin-V/PI双染色流式细胞术观察各组细胞凋亡变化。结果:As2O3与VitC联用能显著降低单用As2O3的细胞存活率(P<0.05),同时VitC能显著增强As2O3诱导细胞凋亡作用(P<0.01)。结论:联用VitC能增强As2O3诱导Hep-2凋亡的作用。
Objective: To investigate the synergistic effect of arsenic trioxide (As2O3) and vitamin C on the apoptosis of Hep-2 cells. Methods: Human Hep-2 cells cultured in vitro were incubated with As2O3 and VitC. Cell viability was determined by MTT assay. Cell apoptosis was observed by flow cytometry with Annexin-V / PI double staining. Variety. Results: As2O3 combined with VitC could significantly reduce the cell viability of As2O3 alone (P <0.05), and VitC could significantly enhance the apoptosis induced by As2O3 (P <0.01). Conclusion: VitC can enhance the apoptosis of Hep-2 induced by As2O3.
