目的比较α-双炔失碳酯(α-Ano)对激素依赖型与非激素依赖型前列腺癌体内外作用强弱的差别。方法应用SRB细胞染色法检测α-Ano对人前列腺癌激素依赖型细胞LNCaP、RV1、L1A和非激素依赖细胞DU145、PC-3生长的影响;裸鼠皮下接种人前列腺癌细胞RV1、PC-3,构建实体瘤模型,成瘤后分组并灌胃给药,观察测量肿瘤体积及质量变化,计算肿瘤增长百分率和抑瘤率,比较α-Ano对激素依赖型和非激素依赖型人前列腺癌裸鼠皮下移植瘤生长的抑制作用。结果α-Ano作用后,LNCaP、RV1和L1A细胞生存率均低于DU145、PC-3细胞;α-Ano高低剂量组均对裸鼠皮下RV1细胞移植瘤具有较强的抑制生长作用,而对PC-3细胞移植瘤作用较弱,仅高剂量组有一定抑制作用。结论α-Ano对激素依赖型前列腺癌的抑制作用强于非激素依赖型前列腺癌。
Objective To compare the effects of α-Ano on the effect of hormone-dependent and non-hormone-dependent prostate cancer in vitro and in vivo. Results After α-Ano treatment, the survival rates of LNCaP, RV1 and L1A cells were lower than that of DU145 and PC-3 cells. Both high and low dose α-Ano groups had a significant inhibitory effect on the growth of transplanted subcutaneous RV1 cells in nude mice, The effect of PC-3 cell xenografts was weak, only high-dose group had a certain inhibitory effect. Conclusion The inhibitory effect of α-Ano on hormone-dependent prostate cancer is stronger than that of non-hormone-dependent prostate cancer.
