目的:研究以HPLC法测定小半夏汤中5-羟甲基糠醛含量的方法。方法:采用Lichrospher C18柱(4.6 mm×150 mm,5μm),以甲醇-水(5∶95)为流动相,体积流量1.0 mL·min-1,波长283 nm,柱温30℃。结果:5-羟甲基糠醛在0.304~2.736μg与峰面积呈现良好的线性关系,r=0.999 9;平均加样回收率(n=5)为96.18%,RSD为1.67%。结论:该法操作简便,结果可靠,重复性好,可作为研究小半夏汤中5-羟甲基糠醛含量的方法。
Objective: To study the HPLC method for the determination of 5-hydroxymethyl furfural in Xiaobanxia Decoction. Methods: The mobile phase consisted of Lichrospher C18 column (4.6 mm × 150 mm, 5 μm) with methanol-water (5:95) as mobile phase at a flow rate of 1.0 mL · min-1 with a wavelength of 283 nm and column temperature of 30 ℃. Results: The linear range of 5-HMF at 0.304 ~ 2.736μg showed a good linear relationship with the peak area (r = 0.999 9). The average recovery was 96.18% (n = 5) and the RSD was 1.67%. Conclusion: The method is simple, reliable and reproducible. It can be used as a method to study the content of 5-hydroxymethylfurfural in Xiaobanxia Decoction.
