多发性骨髓瘤的骨髓细胞置于Dexter长期培养体系中,浆细胞可以长期生长。非贴壁层细胞形成CFU-GM的能力达6周以上,但随着培养时间的延长,CFU-GM的形成活性明显下降。浆细胞产生兔疫球蛋白单克隆特异性在Dexter长期培养体系中仍维持不变,但Ki-67阳性的浆细胞随着培养时间的延长而减少,以上结果说明浆细胞虽然可以在Dexter培养体系中生长并保持单克隆特异性,但此种培养体系不是浆细胞最佳的生长条件。本研究还表明Dexter长期培养不宜作为骨髓净化的手段用于多发性骨髓瘤的自体骨髓移植。
The bone marrow cells of multiple myeloma are placed in a Dexter long-term culture system, and plasma cells can grow for a long time. The ability of non-adherent cells to form CFU-GM was more than 6 weeks, but as the culture time prolonged, the formation activity of CFU-GM decreased significantly. Monoclonal production of plasma proteoglycans in rabbit plasma remains unchanged in Dexter’s long-term culture system, but Ki-67-positive plasma cells decrease as culture time increases. The above results indicate that plasma cells can be in Dexter culture system. While growing and maintaining monoclonal specificity, this culture system is not the best growth condition for plasma cells. This study also showed that long-term culture of Dexter should not be used as a means of bone marrow purification for autologous bone marrow transplantation of multiple myeloma.
