目的 :检测基质金属蛋白酶 3(MMP- 3)在大鼠正常动脉及腹动脉瘤模型组织中的表达 ,以探讨其在腹主动脉瘤发病机制中的作用。方法 :建立大鼠腹主动脉瘤灌注模型 ,应用免疫组织化学和分子原位杂交技术检测 MMP- 3的表达。结果 :正常动脉壁组织 MMP- 3蛋白表达为弱阳性或阴性 ,而动脉瘤组织中 MMP- 3蛋白表达显著增高 ,阳性率为 8/ 8;MMP- 3阳性染色主要位于中膜的平滑肌细胞和外膜的炎症细胞 ,其中炎症细胞浸润的区域染色明显增强。 MMP- 3m RNA在动脉瘤组织中呈阳性表达 ,在动脉外膜炎症细胞浸润的区域和小血管聚集区呈强阳性 ;对照组动脉组织未见 MMP- 3m RNA阳性表达。 结论 :腹主动脉瘤组织中 MMP- 3蛋白及 m RNA较正常动脉组织表达显著增高 ,尤其在炎症细胞浸润的区域表达明显增强 ;提示 MMP- 3可能在腹主动脉瘤发生发展过程中起到关键的起始因子的作用
OBJECTIVE: To detect the expression of matrix metalloproteinase-3 (MMP-3) in the normal and abdominal aneurysm models of rats to investigate its role in the pathogenesis of abdominal aortic aneurysms. Methods: The rat abdominal aortic aneurysm perfusion model was established. The expression of MMP-3 was detected by immunohistochemistry and in situ hybridization. Results: The expression of MMP-3 protein in normal arterial wall tissue was weakly positive or negative, while the expression of MMP-3 protein in aneurysm tissue was significantly increased, with a positive rate of 8/8. MMP-3 positive staining was mainly located in smooth muscle cells and The adventitial inflammatory cells, in which inflammatory cell infiltration area significantly enhanced staining. The expression of MMP-3 mRNA in the aneurysm tissues was strongly positive in the areas infiltrated by adventitial inflammatory cells and in the small blood vessel aggregated area. The MMP-3 mRNA was not found in the control group. CONCLUSION: The expression of MMP-3 protein and m RNA in normal abdominal aortic aneurysm tissues is significantly higher than that in normal artery tissues, especially in the areas infiltrated by inflammatory cells.
