目的研究常见3种食品添加剂联合使用对小鼠生殖腺组织学的影响。方法从方便面和火腿肠联合食用问卷中选出不同品牌使用频率最高的3种食品添加剂,用L16(45)正交表设16个实验组,每组6只成年小鼠,体重18~22g,雌雄各半;设对照组和染毒剂量低、中、高组,剂量为:山梨酸钾250、1 250和2 500 mg/kg,谷氨酸钠100、500和1000mg/kg,亚硝酸钠20、100和200mg/kg;每天定时灌胃染毒1次,连续14d后,取小鼠睾丸和卵巢,经切片染色,显微镜下观察组织学改变。结果各染毒组睾丸结构完整,未见异常变化;卵巢见卵泡构成紊乱,但无论是2因素还是3因素组,对小鼠成熟卵泡的联合毒性作用均未显示统计学意义。结论常见3种食品添加剂联合作用对小鼠生殖腺的组织学影响不明显,未见相应组织学改变。
Objective To study the effect of common three kinds of food additives on gonadal histology in mice. Methods Three kinds of food additives with the highest frequency of different brands were selected from the questionnaire of instant noodles and sausage combined with sixteen experimental groups with L16 (45) orthogonal table. Each group of six adult mice weighing 18-22g, The control group and exposure to low, medium and high doses of potassium sorbate 250,1 250 and 2 500 mg / kg, sodium glutamate 100,500 and 1000mg / kg, sodium nitrite 20, 100 and 200 mg / kg respectively. The mice were orally administered once a day for 14 days. The testis and ovary of mice were stained by sections and the histological changes were observed under microscope. Results The testis structure of each exposure group was intact with no abnormal changes. Ovarian follicles were found to be disorderly formed. However, the joint toxic effects on mature follicles of mice were not statistically significant, no matter in 2-factor or 3-factor groups. Conclusion The common effect of three kinds of food additives on the histology of gonads in mice was not obvious, and no corresponding histological changes were observed.
