目的评价栀子大黄汤(Zhi-Zi-Da-Huang decoction,ZZDHD)抗酒精性肝损伤的药效作用。方法建立体外酒精性大鼠肝细胞损伤模型,考察ZZDHD含药血清对损伤肝细胞的增殖(methylthiazoletrazolium,MTT)及谷丙转氨酶(alanine aminotransferase,ALT)泄漏的影响。建立酒精性大鼠肝损伤体内模型,不同剂量给药后测定血清ALT,谷草转氨酶(aspartate aminotransferase,AST)和甘油三酯(triglyceride,TG)及肝组织中超氧化物歧化酶(superoxide dismutase,SOD),谷胱甘肽(reduced glutathione,GSH)和丙二醛(malondialdehyde,MDA)的含量与活性,并进行肝组织病理学检查。结果体外细胞试验中,体积分数为20%的含药血清能非常显著提高损伤肝细胞的增殖能力并降低细胞ALT泄漏(P<0.01),其活性最佳。大鼠体内,ZZDHD高、中剂量组均能不同程度地降低由酒精诱导的血清ALT、AST和TG水平的升高(P<0.01或P<0.05);均能非常显著性降低受损肝组织中的MDA含量(P<0.01),升高肝组织SOD活力和GSH含量(P<0.01),同时组织病理学也得出上述一致的结论。结论 ZZDHD对酒精性肝损伤具有明显的防治作用,其作用机制可能与减轻自由基和脂质过氧化物对肝细胞的损伤有关。
Objective To evaluate the pharmacodynamic effects of Zhi-Zi-Da-Huang decoction (ZZDHD) on alcohol-induced liver injury. Methods The rat model of alcoholic liver injury was established in vitro and the effects of serum containing ZZDHD on the leakage of methylthiazoletrazolium (MTT) and alanine aminotransferase (ALT) were observed. The model of hepatic injury in alcoholic rats was established in vivo. Serum levels of ALT, AST and triglyceride (TG) and superoxide dismutase (SOD) , Glutathione (GSH) and malondialdehyde (MDA) content and activity, and liver histopathological examination. Results In vitro experiment, the drug-containing serum with a volume fraction of 20% could significantly increase the proliferation of injured hepatocytes and reduce the ALT leakage (P <0.01). In vivo, ZZDHD high and middle dose groups could reduce the level of serum ALT, AST and TG induced by alcohol (P <0.01 or P <0.05) to different extents; both could significantly reduce the damage of impaired liver tissue (P <0.01), and increased SOD activity and GSH content in liver tissue (P <0.01). Meanwhile, the same conclusions were drawn from histopathology. Conclusion ZZDHD has a significant preventive and therapeutic effect on alcoholic liver injury. Its mechanism may be related to the reduction of the damage of hepatocytes by free radicals and lipid peroxides.
