目的应用可变数目串联重复序列(VNTR)分子分型技术,对宁波地区138株肺结核临床分离株进行分型研究,探讨宁波地区结核菌株DNA多态性和基因型特征。方法根据文献选取7个分型效果较好的VNTR基因位点,应用聚合酶链反应(PCR)和琼脂糖凝胶电泳,分析结核分枝杆菌DNA多态性。结果共对138株结核分枝杆菌的7个VNTR位点进行了检测,共产生7个基因簇和123个独立基因型,成簇率为5.8%,VNTR-7位点基因分型技术分辨率指数(HGI)为0.9991,VNTR-3820位点的多态性最高。结论宁波地区的结核分枝杆菌存在较高的基因多态性,且VNTR-7位点基因分型技术有较高的分辨力,适用于宁波地区结核病分子流行病学研究。
Objective To study the genotyping and DNA polymorphism of tuberculosis isolates from 138 isolates of tuberculosis in Ningbo region by VNTR molecular typing technique. Methods According to the literature, seven genotyping VNTR loci were selected, and DNA polymorphisms of Mycobacterium tuberculosis were analyzed by polymerase chain reaction (PCR) and agarose gel electrophoresis. Results A total of 7 VNTR sites of 138 strains of Mycobacterium tuberculosis were detected. Seven gene clusters and 123 independent genotypes were generated. The clustering rate was 5.8%. The VNTR-7 locus genotyping resolution The index (HGI) was 0.9991, with the highest polymorphism at VNTR-3820 locus. Conclusion Mycobacterium tuberculosis in Ningbo region has high genetic polymorphism, and VNTR-7 loci genotyping technique has higher resolution and is suitable for molecular epidemiology of tuberculosis in Ningbo.
