目的:探讨山茱萸药材鲜品与干品总DNA的提取方法,比较鲜、干品总DNA质量,为以后的研究奠定基础。方法:采用改良CTAB法A,B和经典SDS法,提取山茱萸药材鲜品和干品总DNA,对其进行DNA含量测定和电泳检测。结果:改良CTAB法A提取山茱萸药材鲜品与干品总DNA的OD260/OD280比值在1.7~1.9之间,电泳条带清晰,无拖尾,对山茱萸药材鲜品与干品DNA进行综合比较,结果显示鲜品获得的DNA质量高于干品。结论:改良的CTAB法A是分离高质量完整DNA的方法简便、快速,该方法得到的DNA适合下一步分子生物学研究。
Objective: To explore the extraction method of total DNA of fresh and dried products of medicinal materials of Cornus officinalis, and to compare the total DNA quality of fresh and dried products, which lays the foundation for future research. Methods: The total DNA of fresh and dried Cornus officinalis was extracted by improved CTAB method A, B and classical SDS method. The content of DNA was determined and electrophoresed. Results: The ratio of OD260 / OD280 of the total DNA of fresh and dried samples of Codonopsis officinalis extracted by modified CTAB method was between 1.7 and 1.9. The electrophoresis band was clear and without tailing. The DNA of fresh and dried Cornus officinalis was comprehensively compared, The results showed that fresh products obtained higher quality DNA than dried products. Conclusion: The modified CTAB method A is a simple and rapid method for the isolation of high quality intact DNA. The DNA obtained by this method is suitable for further molecular biology research.
