目的研究不同提取路线提取的合欢皮皂苷对荷瘤小鼠的毒性影响。方法取H22肝癌细胞腹腔内接种7 d的小鼠腹水,无菌条件下于小鼠的右腋下接种制备小鼠实体瘤模型。实验分为对照组、模型组、3条不同路线提取的皂苷组(分别记为组分1、组分2、组分3,5 mg.kg?1.d?1)。连续观察10 d,记录小鼠体重、饮食变化、小鼠存活率,并计算其器官指数。结果与模型组相比,皂苷组的小鼠均出现了不同程度的毒性表现,体重和饮食量呈现一定的下降趋势;组分1、组分2、组分3的存活率分别为75%,50%,62.5%;组分1对各器官指数无明显差异,组分2明显增加小鼠的肝、脑器官指数,组分3的皂苷明显增加小鼠的脑器官指数。结论 3条不同路线提取的皂苷对荷瘤小鼠毒性不同,以组分2毒性最大。
Objective To study the toxic effects of sophora saponins extracted from different extraction routes on tumor-bearing mice. Methods Mouse H22 hepatoma cells were inoculated intraperitoneally for 7 days in mice ascites and inoculated to the right armpit of mice under sterile conditions to prepare mouse solid tumor model. The experiment was divided into control group, model group and saponin extracted from three different routes (component 1, component 2, component 3, 5 mg.kg -1, d 1 respectively). After 10 days of continuous observation, the body weight and diet of mice were recorded, the survival rate of mice was recorded, and the organ index was calculated. Results Compared with the model group, the mice in the saponin group showed different degrees of toxicity, and the body weight and diet decreased. The survival rates of component 1, component 2 and component 3 were 75% 50%, and 62.5% respectively. There was no significant difference in component index of each component between component 1 and component 2, and the index of liver and brain organ of component 2 was increased obviously. The saponin of component 3 significantly increased the index of brain organ of mice. Conclusion Saponins extracted from three different routes have different toxicity to tumor-bearing mice, and component 2 has the highest toxicity.
