目的:优选黄芪中免疫活性成分的提取工艺。方法:以黄芪甲苷和黄芪总多糖提取量、干浸膏得率为综合评价指标,通过单因素试验考察水提法、1%NaOH温浸法、1%CaO煎煮法对黄芪中免疫活性成分提取效率的影响。采用HPLC测定黄芪甲苷含量,流动相乙腈-水(30∶70),空气泵压力0.4 MPa,ELSD漂移管温度105℃,载气流速2.7 L·min-1;利用UV测定黄芪总多糖含量,检测波长489 nm。结果:3种提取方式的黄芪甲苷提取量分别为0.826,1.016,0.842 mg·g-1,黄芪总多糖提取量分别为14.49,5.82,17.04 mg·g-1,干浸膏得率无显著性差异。结论:采用水提法能达到较好的提取效果,同时生产成本较低,适用于黄芪中免疫活性成分的工业大生产。
Objective: To optimize the extraction process of immunocompetent ingredients in Radix Astragali. Results: The extraction rates of Astragaloside Ⅳ, Astragaloside Ⅳ and Astragalus membranaceus were 0.826, 1.016 and 0.842 mg · g-1, respectively, and the total polysaccharides of Astragalus were 14.49, 5.82 and 17.04 mg · g-1, respectively. Sex differences. Conclusion: The water extraction method can achieve better extraction results, while the production cost is lower, suitable for industrial production of immunologically active ingredients in Astragalus.
