目的:研究原癌基因C-myb反义寡核苷酸在裸鼠体内诱导脑胶质瘤细胞凋亡及抑制生长的作用。方法:采用人工合成反义、正义二组硫代脱氧寡核酸(SON),C6脑胶质瘤瘤体内直接注入法,用游标卡尺检测肿瘤的生长抑制率,HE染色方法检测细胞的凋亡状况。结果:C-myb反义SON组脑胶质瘤细胞的生长明显减慢,瘤细胞凋亡增加。结论:C-myb反义SON能够诱导C6脑胶质瘤细胞的凋亡,激发脑胶质瘤细胞的特异性免疫反应,同时能明显抑制肿瘤细胞的生长。
Objective: To study the effect of C-myb antisense oligodeoxynucleotide on the apoptosis of glioma cells in nude mice and its inhibition of growth. Methods: Synthesized antisense and sense group thiodeoxyl oligodeoxynucleotides (SON) and C6 glioma tumors were directly injected in vivo. The tumor growth inhibition rate was detected by vernier caliper. HE staining was used to detect the cell apoptosis. RESULTS: The growth of C-myb antisense glioma cells was significantly slower and the apoptosis of tumor cells increased. Conclusion: C-myb antisense SON can induce apoptosis of C6 glioma cells, stimulate the specific immune response of glioma cells, and can significantly inhibit the growth of tumor cells.
