目的:研究加味四逆散对慢性不可预计性刺激致抑郁雌性大鼠血清皮质酮(Cort)、血清雌二醇(E2)的影响,揭示其对雌性大鼠抑郁模型内分泌紊乱的调节机制。方法:Wistar大鼠按体重和基础糖水消耗量随机分为6组:空白组(不施加任何刺激)、慢性轻度不可预计应激模型组、加味四逆散高、中、低剂量组(315,158,78 mg.kg-1,ig),阳性药氟西汀组(18 mg.kg-1,ig),应激第1天开始每天8:30 ig给药,正常组及慢性应激模型组每次ig蒸馏水。应激第22天取材。放射免疫法检测用药前后雌性大鼠血清Cort,E2的含量变化。结果:大鼠血清Cort空白组(0.05±0.04)μg.L-1,模型组为(0.11±0.08)μg.L-1,较空白组显著升高(P<0.05),加味四逆散各组Cort较模型组低但差异无统计学意义;血清E2含量模型组大鼠的E2水平(0.04±0.02)μg.L-1较空白组(0.12±0.03)μg.L-1明显下降(P<0.05),加味四逆散高、中、低剂量组E2依次为(0.10±0.04),(0.08±0.02),(0.07±0.05)μg.L-1均明显升高,与模型组相比有显著性差异(P<0.01,P<0.05)。结论:加味四逆散抗抑郁作用与其维持E2正常分泌有关。
Objective: To investigate the effects of Jiawei Sini Decoction on serum corticosterone (Cort) and serum estradiol (E2) in female rats induced by chronic unpredictable depression, and to reveal the regulatory mechanism of Jiawei Sini Decoction on endocrine disorders in female rats with depression model. Methods: Wistar rats were randomly divided into 6 groups according to body weight and basic sugar consumption: blank group (without any stimulation), chronic mild unpredictable stress model group, Jiawei Sini Powder high, medium and low dose groups (315,158 , 78 mg.kg-1, ig) and positive drug fluoxetine (18 mg.kg-1, ig). The rats in the normal group and the chronic stress model group Distilled water every ig. Stress the first 22 days drawing. Radioimmunoassay was used to detect the changes of serum Cort and E2 levels in female rats before and after treatment. Results: Compared with the blank group, the level of serum Cort (0.05 ± 0.04) μg.L-1 in model group was (0.11 ± 0.08) μg.L-1, Group Cort was lower than the model group, but the difference was not statistically significant. The level of E2 in the serum E2 group was significantly lower than that in the blank group (0.04 ± 0.02) μg.L-1 (P 0.12 ± 0.03) μg.L-1 <0.05). The E2 of Jiawei Sini San high, medium and low dose groups were (0.10 ± 0.04), (0.08 ± 0.02) and (0.07 ± 0.05) μg.L-1, respectively There was significant difference (P <0.01, P <0.05). Conclusion: The anti-depressive effect of Jiawei Sini Decoction is related to maintaining the normal secretion of E2.
