目的 研究VEGF -C在人舌癌发生发展中的作用 ,检测VEGF -CmRNA和蛋白在人舌鳞癌细胞株TCa8113中的表达 ,建立高表达VEGF -C的细胞株TCa8113 /VEGF -C。方法 运用RT -PCR和免疫组化法分别检测VEGF -CmRNA和蛋白在人舌癌细胞株中的表达 ,通过脂质体转基因的方法筛选建立过表达VEGF-C的细胞株。结果 TCa8113中检测到VEGF -C的表达 ,筛选建立的细胞株高表达VEGF -C。结论 TCa8113能转录VEGF -CmRNA ,并在胞浆中翻译表达VEGF -C蛋白 ,但是表达较弱 ;筛选建立的高表达VEGF-C的TCa8113细胞株为今后的研究提供了材料
Objective To investigate the role of VEGF-C in the development of human tongue cancer, and to detect the expression of VEGF-C mRNA and protein in human tongue squamous cell carcinoma cell line TCa8113 and to establish the cell line TCa8113 / VEGF-C highly expressing VEGF-C. Methods The expression of VEGF-C mRNA and protein in human tongue cancer cell lines was detected by RT-PCR and immunohistochemistry. The cell lines overexpressing VEGF-C were screened by liposome transgene method. Results The expression of VEGF-C was detected in TCa8113 cells. The established cell lines highly expressed VEGF-C. CONCLUSION TCa8113 can transcribe VEGF-C mRNA and express VEGF-C protein in the cytoplasm, but its expression is weak. The TCa8113 cell line screened for high expression of VEGF-C provides material for future research
