目的观察在不同浓度脂多糖(LPS)干预下,热休克蛋白27(Hsp27)在人牙龈成纤维细胞(HGFs)损伤过程中的表达。方法取原代培养并鉴定3~8代的HGFs,采用细胞划痕实验检测不同浓度LPS对HGFs体外迁移的影响;以LPS梯度浓度干预,采用Western blotting方法检测Hsp27在HGFs中的表达。结果 LPS浓度越高细胞迁移能力越弱;Hsp27在正常HGFs中呈弱阳性表达,在LPS干预后的HGFs中呈强阳性表达,且随干预浓度增高表达量有逐渐增高的趋势,与细胞迁移能力呈反比。结论 Hsp27在LPS介导的HGFs损伤中表达量升高,提示Hsp27在LPS介导的HGFs损伤过程中具有重要作用。
Objective To observe the expression of heat shock protein 27 (Hsp27) in human gingival fibroblasts (HGFs) under the intervention of different concentrations of lipopolysaccharide (LPS). Methods HGFs were cultured and identified from 3 to 8 generations in primary culture. The effects of different concentrations of LPS on the migration of HGFs in vitro were detected by cell scratch assay. The expression of Hsp27 in HGFs was detected by Western blotting with LPS gradient concentration. Results The higher the concentration of LPS was, the weaker the cell migration ability. The expression of Hsp27 was weakly positive in normal HGFs and strongly positive in HGFs after LPS intervention. The expression level of Hsp27 was gradually increased with the increase of intervention concentration, Inversely. Conclusion The expression of Hsp27 is up-regulated in LPS-induced HGFs injury, suggesting that Hsp27 plays an important role in LPS-mediated HGFs injury.
