目的探讨雷公藤甲素对新生大鼠心肌细胞搏动的影响。方法新生大鼠心肌细胞予0、2.5、5、10、20、40、80、160和200μmol/L雷公藤甲素处理,采用实时细胞分析系统(RTCA)于给药前至给药后20 h连续记录心肌细胞搏动频率的变化。结果浓度大于等于2.5μmol/L的雷公藤甲素于给药后10 min可明显导致心肌细胞搏动频率明显下降,但1h后可基本恢复至给药前水平,给药后1h至给药后10 h,细胞搏动频率未见明显异常变化,心肌细胞搏动节律正常。160和200μmol/L组给药后20 h,细胞搏动频率明显下降,正常节律性搏动消失。结论较高浓度的雷公藤甲素给药后期对大鼠心肌细胞毒性作用显著。
Objective To investigate the effect of Triptolide on cardiomyocyte pulsatility in neonatal rats. Methods The neonatal rat cardiomyocytes were treated with 0, 2.5, 5, 10, 20, 40, 80, 160 and 200 μmol / L triptolide using real time cell analysis system (RTCA) Continuous recording of changes in cardiomyocyte pulsation frequency. Results Triptolide at a concentration of 2.5μmol / L or more significantly reduced the frequency of cardiomyocytes pulsation 10 min after administration, but returned to the pre-administration level after 1h, and after 1h to 10 h, there was no obvious abnormality in the frequency of cell beating, and the beating rhythm of cardiomyocytes was normal. At 20 h after administration of 160 and 200 μmol / L groups, the frequency of cell pulsation was significantly decreased and the normal rhythmic beat disappeared. Conclusion Triptolide at a higher concentration has a significant cytotoxic effect on rat myocardial cells at the later stage of administration.
