目的观察吸烟大鼠脑动脉内皮细胞P-选择素、E-选择素表达,探讨吸烟致脑血栓形成的机制。方法建立吸烟动物模型,将50只大鼠随机分为正常对照组,短期大量吸烟组,长期小量吸烟组,长期大量吸烟组,戒烟组。免疫组织化学法检测大鼠脑动脉内皮细胞P-选择素、E-选择素和TNF-α的表达。结果吸烟大鼠脑血管内皮细胞P-选择素、E-选择素和TNF-α表达较不吸烟鼠明显增加(P<0.05)。长期大量吸烟组P-选择素、E-选择素和TNF-α表达比其他各组明显增多,戒烟组P-选择素、E-选择素和TNF-α表达下降,与其他各吸烟组相比有显著性差异(P<0.05)。结论吸烟致大鼠脑血管内皮细胞P-选择素、E-选择素和TNF-α表达升高是脑血栓形成的重要机制之一。
Objective To observe the expression of P-selectin and E-selectin in cerebral artery of smoking rats and explore the mechanism of cerebral thrombosis caused by smoking. Methods A smoking animal model was established. Fifty rats were randomly divided into normal control group, short-term heavy smoking group, long-term small smoking group, long-term heavy smoking group and smoking cessation group. Immunohistochemistry was used to detect the expression of P-selectin, E-selectin and TNF-α in rat cerebral arterial endothelial cells. Results Compared with non-smoking rats, the expression of P-selectin, E-selectin and TNF-α in smoking rat cerebrovascular endothelial cells were significantly increased (P <0.05). The expressions of P-selectin, E-selectin and TNF-α in long-term heavy smoking group were significantly higher than those in other groups. The expressions of P-selectin, E-selectin and TNF-αwere decreased in smoking cessation group compared with other smoking groups There was significant difference (P <0.05). Conclusions The increased expression of P-selectin, E-selectin and TNF-α in rat cerebrovascular endothelial cells induced by smoking is one of the important mechanisms of cerebral thrombosis.
