流式细胞术检测宫颈癌CD105与VEGF-C、nm23表达

目的:探讨血管内皮生长因子-C(vascular endothelial growth factor-C,VEGF-C)和nm23(non-metastasis23)表达与CD105及宫颈癌转移的关系。方法:采用免疫组化SABC法检测16例正常宫颈上皮及68例宫颈癌中VEGF-C、nm23的表达,并用流式细胞术检测内皮细胞CD105阳性率。结果:VEGF-C在宫颈癌组织中的阳性表达率显著高于正常组织(P<0.01),淋巴结转移组VEGF-C阳性率高于无淋巴结转移组(P<0.05),VEGF-C表达强度与CD105阳性率呈正相关(P<0.05)。nm23蛋白表达率显著低于正常组织(P<0.01),淋巴结转移组nm23阳性率低于无淋巴结转移组(P<0.05),nm23在宫颈癌中的表达与VEGF-C呈负相关(P<0.01)。结论:宫颈癌组织VEGF-C表达升高、nm23低表达是血管新生、肿瘤浸润转移过程的关键环节;CD105高表达与子宫颈癌浸润转移密切相关。 Objective: To investigate the relationship between the expression of vascular endothelial growth factor-C (VEGF-C) and nm23 (non-metastasis23) and the metastasis of cervical cancer. Methods: Immunohistochemical SABC method was used to detect the expression of VEGF-C and nm23 in 16 cases of normal cervical epithelium and 68 cases of cervical cancer. Flow cytometry was used to detect the positive rate of CD105 in endothelial cells. Results: The positive rate of VEGF-C in cervical cancer was significantly higher than that in normal tissues (P <0.01). The positive rate of VEGF-C in lymph node metastasis was higher than that in non-lymph node metastasis (P <0.05) And the positive rate of CD105 was positively correlated (P <0.05). The positive rates of nm23 in patients with lymph node metastasis were significantly lower than those without lymph node metastasis (P <0.05), while the expressions of nm23 in patients with cervical cancer were negatively correlated with VEGF-C (P < 0.01). Conclusion: The expression of VEGF-C is increased in cervical cancer tissues. The low expression of nm23 is a key link in the process of angiogenesis and tumor invasion and metastasis. The high expression of CD105 is closely related to the invasion and metastasis of cervical cancer.
