采用体外细胞培养的方法 ,观察复合真菌多糖诱导人肝癌细胞 Bel- 740 2凋亡的影响 ,利用双重免疫标记方法分析肿瘤抑制基因 P5 3在 Bel- 740 2凋亡中的表达。结果表明 :不同浓度的复合真菌多糖均能明显诱导 Bel- 740 2细胞凋亡 ,并随浓度增加而渐次出现不同时相的细胞凋亡特征性染色质凝聚 ;能明显降低G1 、S期 ,且有显著的剂量 -效应关系 ,凋亡细胞百分率亦随浓度的增加而逐渐升高 ;在高浓度组出现典型的DNA阶梯状电泳条带。证明复合真菌多糖抗肿瘤作用机理与诱导肿瘤细胞的凋亡密切相关 ,其诱导肿瘤细胞凋亡是 P5 3非依赖性。
The in vitro cell culture method was used to observe the effect of compound fungal polysaccharide on the apoptosis of human hepatocellular carcinoma cell Bel-740 2 . The double immunolabeling method was used to analyze the expression of tumor suppressor gene P53 in Bel-7402 apoptosis. The results showed that different concentrations of complex fungal polysaccharides could induce apoptosis of Bel-7402 cells, and with the increase of concentration, there were gradually different characteristics of chromatin condensation at different phases of apoptosis, and G1 and S phases could be significantly decreased. There was a significant dose-effect relationship, and the percentage of apoptotic cells also increased gradually with increasing concentrations; a typical DNA ladder was observed in the high concentration group. It is proved that the anti-tumor mechanism of the complex fungal polysaccharide is closely related to the induction of apoptosis in tumor cells, and its induction of tumor cell apoptosis is P5 3 independent.
