目的研究蛋白酶体抑制剂MG132和顺铂的联合用药前后卵巢癌细胞株Caov-3细胞内NF-κB和cyclinD1的表达变化情况。方法免疫组织化学SP法测定用药前后Caov-3细胞中NF-κB和cyclinD1的表达情况。MTT法检测细胞的生长抑制情况。结果免疫组织化学结果显示MG132和顺铂联合用药作用24h后Caov-3细胞中NF-κB和cyclinD1的表达下调,与对照组和顺铂组比较差异有显著性(P<0.01),联合用药组中不同MG132浓度的各组间两两比较差异有显著性(P<0.01),联合用药能显著增加细胞的生长抑制率(P<0.01)。结论 MG132能诱导卵巢癌细胞Caov-3的凋亡,其重要的作用机制可能是通过抑制NF-κB和cyclinD1的表达来抑制肿瘤细胞的生长和诱导肿瘤细胞的凋亡。
Objective To investigate the expression of NF-κB and cyclinD1 in ovarian cancer cell line Caov-3 before and after the combination of proteasome inhibitor MG132 and cisplatin. Methods The expressions of NF-κB and cyclinD1 in Caov-3 cells before and after treatment were measured by immunohistochemical SP method. MTT assay of cell growth inhibition. Results The results of immunohistochemistry showed that the expressions of NF-κB and cyclinD1 were down-regulated in Caov-3 cells treated with MG132 and cisplatin for 24 h, which were significantly lower than those in control group and cisplatin group (P <0.01) (P <0.01). The combined treatment could significantly increase the cell growth inhibition rate (P <0.01). Conclusion MG132 can induce the apoptosis of ovarian cancer cells Caov-3. The possible mechanism is that MG132 can inhibit the growth of tumor cells and induce the apoptosis of tumor cells by inhibiting the expression of NF-κB and cyclinD1.
