目的研究大黄素对脓毒症大鼠高血糖及胰岛素抵抗的影响。方法 60只SD大鼠随机分为假手术组、模型组、大黄素组、二甲双胍阳性对照组。手术前1 h,大黄素组灌胃给予100 mg·kg~(-1)的CMC-Na混悬液,二甲双胍阳性对照组灌胃给予150 mg·kg-1的CMC-Na混悬液,按照盲肠结扎穿刺法建立脓毒症模型。分别测定术前及术后1,2,4,6,12,24 h的血糖值,及术后2 h的胰岛素水平,计算胰岛素抵抗指数,并于术后24 h腹主动脉取血,酶联免疫吸附法检测IL-6、TNF-α、CRP水平。结果大黄素能改善脓毒症大鼠的胰岛素抵抗,抑制脓毒症大鼠感染后血糖的升高,并能明显降低炎症因子IL-6、TNF-α、CRP的水平。结论大黄素能降低脓毒症大鼠的高血糖,改善其胰岛素抵抗,其作用机制可能与抑制炎症细胞因子分泌有关。
Objective To study the effects of emodin on hyperglycemia and insulin resistance in septic rats. Methods Sixty SD rats were randomly divided into sham operation group, model group, emodin group and metformin positive control group. One hour before operation, the emodin group was orally administered with 100 mg · kg -1 of CMC-Na suspension. The metformin positive control group was orally administered with 150 mg · kg -1 of CMC-Na suspension. Cecal ligation and puncture method to establish sepsis model. The blood glucose levels at 1, 2, 4, 6, 12, and 24 hours after operation and the levels of insulin at 2 hours after operation were measured respectively. Insulin resistance index was calculated and blood was taken from abdominal aorta 24 hours after operation. The levels of IL-6, TNF-α and CRP were detected by ELISA. Results Emodin could improve insulin resistance in septic rats, inhibit the increase of blood glucose in septic rats and significantly decrease the levels of inflammatory cytokines IL-6, TNF-α and CRP. Conclusion Emodin can reduce hyperglycemia and improve insulin resistance in sepsis rats, and its mechanism may be related to the inhibition of the secretion of inflammatory cytokines.
