目的:观察复方三金胶囊对人乳腺癌(MCF-7)细胞生长的抑制作用并探讨其作用机制。方法:采用MTT法检测复方三金胶囊对MCF-7细胞增殖的抑制作用,流式细胞仪检测复方三金胶囊对MCF-7细胞凋亡和周期的影响,Western-blot法检测复方三金胶囊对MCF-7细胞中凋亡相关蛋白PLK1的表达。结果:复方三金胶囊对MCF-7细胞增殖有明显抑制作用,IC50值为390.38μg·ml-1;复方三金胶囊诱导细胞凋亡作用明显,并呈现明显的量效关系;复方三金胶囊对细胞周期的影响主要表现在低浓度时为S期阻滞,高浓度时为G0/G1期阻滞;复方三金胶囊可明显抑制MCF-7细胞中PLK1蛋白的表达。结论:复方三金胶囊体外实验可明显抑制MCF-7细胞的增殖,其抗肿瘤作用与诱导细胞凋亡,改变细胞周期分布,抑制PLK1蛋白表达有关。
Objective: To observe the inhibitory effect of Fufang Sanjin Capsule on the growth of human breast cancer (MCF-7) cells and to explore its mechanism. Methods: The inhibitory effect of Fufang Sanjin Capsule on the proliferation of MCF-7 cells was detected by MTT assay. The effect of Fufang Sanjin capsule on apoptosis and cycle of MCF-7 cells was detected by flow cytometry. The effect of Fufang Sanjin Capsule On the expression of apoptosis-related protein PLK1 in MCF-7 cells. Results: Fufang Sanjin capsule could significantly inhibit the proliferation of MCF-7 cells with the IC50 value of 390.38μg · ml-1. The effect of Fufang Sanjin capsule on apoptosis was obvious and the dose-effect relationship was obvious. Fufang Sanjin capsule The effect on cell cycle was mainly in S phase arrest at low concentration and G0 / G1 arrest at high concentration. Fufang Sanjin Capsule could significantly inhibit the expression of PLK1 protein in MCF-7 cells. CONCLUSION: Fufang Sanjin capsule can significantly inhibit the proliferation of MCF-7 cells in vitro. The anti-tumor effect of Fufang Sanjin capsule can induce the apoptosis, change the cell cycle distribution and inhibit the expression of PLK1 protein.
