目的检测口腔扁平苔藓黏膜上皮下结缔组织中γ-干扰素(IFN-γ)、白细胞介素-10(IL-10)的表达情况,初步探讨其在口腔扁平苔藓发生发展过程中的作用及意义。方法应用PCR技术和荧光实时定量PCR技术检测11例口腔扁平苔藓黏膜上皮下结缔组织和5例正常口腔黏膜上皮下结缔组织中IFN-γ、IL-10的表达情况。结果荧光实时定量PCR检测结果显示,口腔扁平苔藓黏膜上皮下结缔组织中IFN-γ mRNA的表达值实验组与对照组分别为0.2981±0.2064和0.0986±0.0894,扁平苔藓组织均高于正常口腔黏膜组织;IL-10 mRNA的表达值实验组与对照组分别为4.3218±4.4678和15.1156±9.1392,扁平苔藓组织均低于正常口腔黏膜组织,经检验差异有统计学意义(P<0.05)。结论与正常口腔黏膜组织相比,口腔扁平苔藓组织中具有促炎作用的IFN-γ表达水平增高,而具有抗炎作用的IL-10表达水平降低,提示抗炎症反应和促炎症反应在口腔扁平苔藓的发生发展过程中起到一定的作用。
Objective To detect the expression of IFN-γ and IL-10 in the subepithelial connective tissues of oral lichen planus and to explore the role and significance of the expression of interleukin-10 (IL-10) in the development of oral lichen planus . Methods The expression of IFN-γ and IL-10 in 11 cases of oral lichen mucosa subepithelial connective tissue and 5 cases of normal oral mucosa subepithelial connective tissue were detected by PCR and real-time fluorescence quantitative PCR. Results The results of real-time fluorescence quantitative PCR showed that the expression of IFN-γ mRNA in subepithelial connective tissue of oral lichen planus was 0.2981 ± 0.2064 and 0.0986 ± 0.0894, respectively, which were higher than that of normal oral mucosa . The expression of IL-10 mRNA in experimental group and control group were 4.3218 ± 4.4678 and 15.1156 ± 9.1392, respectively, which were lower than that in normal oral mucosa tissues (P <0.05). Conclusion Compared with normal oral mucosa tissue, the level of IFN-γ in the oral lichen planus tissues is increased, while the expression of IL-10 with anti-inflammatory effect is decreased, suggesting that the anti-inflammatory and pro-inflammatory responses may play an important role in oral flat The occurrence and development of moss play a role.
