通过新生猪缺或缺血性脑损伤模型,检测了脑皮质组织钙、钙调素含量以及磷酸二脂酶活性的变化,探讨在缺氧缺血性脑损伤过程中以上三指标的改变.方法 采用改进的Thompson CaM激活PDE间接测定法检测CaM,反应液中除去工具酶PDE及CaM标准液检测PDE,利用火焰原子吸收分光光度法检测Ca~(2+).结果 脑损伤后单位体积湿重脑组织含钙量变化不明显,钙调素含量及磷酸二脂酶活性分别在损伤后即刻及12小时明显升高.结论 缺氧缺血性脑损伤早期就有钙调素含量的升高,继而引起其靶酶活性升高,造成神经系统的损害.
To detect the changes of calcium, calmodulin and phosphodiesterase activity in cortex of newborn pigs with ischemic or ischemic brain injury model and to explore the changes of the above three indexes in the process of hypoxic-ischemic brain damage.Methods CaM was detected by modified Thompson CaM-activated PDE indirect assay, PDE and CaM standard solution were removed from the reaction solution to detect PDE, and Ca 2+ was detected by flame atomic absorption spectrophotometry.Results After wet injury, There was no obvious change of calcium content in brain tissue, and calmodulin content and phosphodiesterase activity increased significantly immediately and 12h after injury.Conclusion The content of calmodulin is increased early in hypoxic-ischemic brain damage, Which in turn leads to its target enzyme activity increased, resulting in nervous system damage.
