以体外组织块培养的方法成功地建立了三个人眼小梁细胞系标本。在倒置显微镜下观察了三代小梁细胞的生长过程,用透射和扫描电子显微镜观察了第三代融合的小梁细胞的超微结构。小梁细胞的生长潜伏期约为1周,原代和传代融合的小梁细胞呈单层细胞生长,单个细胞呈长而扁平状,核位于中央为椭圆形,细胞表面大部分平滑,偶有微绒毛突出,在某些部位可见邻近细胞和其胞突间紧密连在一起。另外,这些细胞还具有高代谢细胞的超微结构特征包括大量的线粒体,发达的Golgi器,丰富的粗面内质网和明显的溶酶体。人小梁细胞体外培养为进一步研究小梁细胞的功能提供了非常有价值的实验物质基础,还可作为青光眼发病学和药物筛选等方面实验研究的基础。
Three human trabecular meshwork cells were successfully established by in vitro tissue culture. The growth of three generations of trabecular cells was observed under an inverted microscope. The ultrastructure of the third generation of trabecular meshwork cells was observed by transmission and scanning electron microscopy. The incubation period of trabecular cells was about 1 week. Primary and subcultured trabecular cells showed monolayer growth. Single cells were long and flat, the nucleus was oval in the center, most of the cell surface was smooth, Fluff prominent, visible in some parts of the neighboring cells and their closely linked together between the cell. In addition, these cells also have ultrastructural features of high-metabolizing cells including a large number of mitochondria, well-developed Golgi apparatus, abundant rough endoplasmic reticulum and significant lysosomes. The in vitro culture of human trabecular cells provides a very valuable experimental material basis for the further study of the function of trabecular cells, and can be used as the basis for experimental studies on the pathogenesis and drug screening of glaucoma.
