目的:研究大孔树脂分离纯化槐花散总黄酮的的工艺条件和参数。方法:采用紫外分光光度法测定总黄酮含量,选择了3种大孔树脂和1种聚酰胺树脂,以槐花散总黄酮吸附率和解吸率为指标,筛选出较优的树脂,并对分离纯化的主要影响因素进行考查,优化工艺条件。结果:AB-8大孔树脂对槐花散总黄酮有良好的纯化性能,适宜的工艺条件为:上样药液浓度为1.5mg/m L,上样体积为2BV,先用4BV水洗,再用3BV 65%乙醇洗脱,树脂径高比为1∶9。结论:AB-8大孔树脂分离纯化槐花散总黄酮效果好、工艺稳定。
Objective: To study the technological conditions and parameters for the separation and purification of total flavonoids from Sophora japonica by macroporous resin. Methods: The total flavonoids were determined by UV spectrophotometry. Three kinds of macroporous resins and one kind of polyamide resin were selected. The optimal resin was screened by the adsorption and desorption rates of total flavonoids of Sophora flavescens. Purification of the main influencing factors to examine and optimize the process conditions. Results: AB-8 macroporous resin had good purification performance on total flavonoids of Sophora flavescens. The optimum technological conditions were as follows: the concentration of sample solution was 1.5mg / m L, the volume of sample was 2BV, first washed with 4BV water, then Eluted with 3BV 65% ethanol, resin diameter ratio of 1: 9. Conclusion: AB-8 macroporous resin separation and purification of total flavonoids from Sophora flavescens has good effect and stable technology.
