目的:运用Box-Behnken响应面法优化翻白草中总黄酮的提取工艺。方法:在单因素试验基础上,以提取时间、乙醇浓度以及料液比为考察因素,以翻白草总黄酮提取率为评价指标,利用Box-Behnken响应面法优化翻白草中总黄酮的提取工艺。结果:提取的最佳工艺为:提取浓度为70%的乙醇,每次120 min,液料比为8∶1 ml·g-1时,黄酮的提取率最高,总黄酮提取率预测值为3.86%,测定值为3.74%,其间相差0.12%。结论:使用Box-Behnken响应面法优化翻白草总黄酮的提取工艺科学可靠,可用于翻白草总黄酮的提取生产。
OBJECTIVE: To optimize the extraction process of total flavonoids from Radix alba by using Box-Behnken response surface methodology. Methods: On the basis of single factor test, taking the extraction time, the ethanol concentration and the solid-liquid ratio as the investigation factors, the extraction rate of total flavonoids of Pseudotsuga rubra was used as evaluation index, and the content of total flavonoids in Pseudostellaria purpurea was optimized by Box-Behnken response surface method Extraction process. Results: The optimum conditions for the extraction were as follows: extraction of 70% ethanol for 120 min, the ratio of liquid to material of 8: 1 ml · g-1, the highest extraction rate of flavonoids, the total flavonoid extraction rate of 3.86 %, Found 3.74%, with a difference of 0.12%. Conclusion: The Box-Behnken response surface method to optimize the extraction process of total flavonoids of Plum is scientific and reliable, which can be used for the extraction of total flavonoids from Plum.
