本文旨在建立简单、快速分析茯苓多糖中单糖组成的高效毛细管电泳法。将茯苓多糖水解,经PMP衍生化,采用高效毛细管电泳分析。电泳条件:缓冲体系为80mmol/L硼砂溶液(pH9.5);毛细管柱(25μm×42cm),分离电压18kV,检测波长254nm,进样高度10cm,进样时间25s。结果显示,茯苓多糖中含葡萄糖、甘露糖和半乳糖的物质的量之比为2.657∶1.000∶2.362。该方法具有简单、快速、准确、重现性好,可用于测定茯苓多糖中单糖的组成。
This paper aims to establish a simple and rapid analysis of the polysaccharides in the polysaccharides of high performance capillary electrophoresis. The tuckahoe polysaccharide hydrolysis, the PMP derivatization, using high performance capillary electrophoresis analysis. Electrophoresis conditions: buffer system is 80mmol / L borax solution (pH9.5); capillary column (25μm × 42cm), separation voltage 18kV, detection wavelength 254nm, injection height 10cm, injection time 25s. The results show that the ratio of the amount of substances containing glucose, mannose and galactose in the tuckahoe polysaccharide is 2.657: 1.000: 2.362. The method is simple, rapid, accurate and reproducible, and can be used to determine the composition of monosaccharides in the tuckahoe polysaccharide.
