目的 研究 5 脱氧杂氮胞苷对肝癌细胞株p16基因表达的影响及其机制。方法 肝癌细胞株SMMC 72 2 1、HePG2 用 5 脱氧杂氮胞苷处理 ,处理前后采用RT PCR和免疫组织化学染色法观察和分析其mRNA和蛋白表达变化。结果 经 5 脱氧杂氮胞苷处理的肝癌细胞株mRNA和蛋白表达均明显增强。结论 5 脱氧杂氮胞苷可使p16基因在转录水平和翻译水平表达增高 ,其机制可能是通过去甲基化使p16基因表达恢复
Objective To investigate the effect of 5-aza cytidine on the expression of p16 gene in hepatoma cell lines and its mechanism. Methods Hepatoma cell line SMMC 72 1 and HePG2 were treated with 5-deoxyazacytidine. RT-PCR and immunohistochemical staining were used to observe and analyze the changes of mRNA and protein expression before and after treatment. Results The mRNA and protein expression of hepatoma cell lines treated with 5-deoxyazacytidine were significantly increased. Conclusion 5-deoxyazacytidine can increase the expression of p16 gene at transcriptional and translational levels. The mechanism may be that demethylation can restore the expression of p16 gene.
