目的:研究β-雌二醇(E2)对去卵巢小鼠子宫内膜组织中Hes-1基因表达水平的影响。方法:将16只雌性昆明系(KM)小鼠随机平均分为卵巢切除生理盐水干预组(OVX+NS)和卵巢切除β-雌二醇干预组(OVX+E2);采用Trizol一步法抽提内膜组织总RNA,进行RT-PCR实验,扩增出Hes-1基因和内参基因(GAPDH)目标条带,采用QuantityOne-4.6.2凝胶分析软件对Hes-1基因与GAPDH基因的目标带进行半定量分析。结果:OVX+NS组和OVX+E2组小鼠子宫内膜组织中Hes-1基因mRNA的相对表达强度分别为:0.16±0.06和0.06±0.01,P<0.05,具有统计学差异。结论:高剂量的β-E2可能通过下调Hes-1基因的表达促进子宫内膜上皮细胞增生,Hes-1基因和ER活性状态之间的相互作用可能在子宫内膜疾病的发展机制中发挥了重要作用。
Objective: To study the effect of β-estradiol (E2) on the expression of Hes-1 gene in endometrium of ovariectomized mice. Methods: 16 female Kunming (KM) mice were randomly divided into ovariectomized NS group (OVX + NS) and ovariectomized group (OVX + E2). Trizol single-step extraction The total RNA of endometrial tissue was amplified by RT-PCR. The target band of Hes-1 gene and internal reference gene (GAPDH) was amplified. Results: The relative expression intensity of Hes-1 mRNA in OVX + NS group and OVX + E2 group was 0.16 ± 0.06 and 0.06 ± 0.01, respectively, with statistical significance. Conclusion: High-dose β-E2 may promote the proliferation of endometrial epithelial cells by down-regulating the expression of Hes-1 gene.
