对10例肾移植患者作山地明转换Neoral试验,行药效动力学对比研究。30例新移植患者应用Neoral进行临床观察。结果,Neoral最高血浓度较山地明高282±84,达峰时间短0.79±0.29,CsA暴露大1533±169(P<0.05)。Neoral有较好的抗排斥作用.但由于吸收迅速,血药浓度显超过治疗窗水平,引起肝中毒达20%。因此,我们认为Neoral与山地明转换,比例应用1:0.8~1:0.9,以免毒副作用发生。本组观察36月,Neoral服药量可较山地明常规剂量减少25%。
In 10 cases of renal transplant patients with mountain Ming conversion Neoral test, the pharmacodynamics comparative study. Thirty new cases of patients were treated with Neoral for clinical observation. The results showed that the highest Neoral blood concentration was 282 ± 84, and the peak time was 0.79 ± 0.29 and 1533 ± 169, respectively (P <0.05). Neoral has a good anti-rejection effect. However, due to rapid absorption, blood concentration significantly over the therapeutic window level, causing liver poisoning up to 20%. Therefore, we think Neoral and mountain Ming conversion, the ratio of 1: 0.8 ~ 1: 0.9, so as to avoid toxic side effects. This group observed 36 months, Neoral dosage can be reduced compared with conventional mountain dose of 25%.
