目的探讨人外周血内皮细胞与大鼠胰岛共移植到糖尿病裸鼠肾包膜下改善移植胰岛存活及其功能发挥情况。方法将人外周血内皮细胞(5×105个)与新鲜分离的大鼠胰岛细胞共移植到糖尿病裸鼠模型肾包膜下,检测移植受体裸鼠空腹血糖水平、体质量和外周血C肽水平,并行腹腔葡萄糖耐量试验(IPGTT),以了解移植胰岛的存活及其功能情况。结果与对照组相比,实验组受体的血糖明显下降(P<0.01),外周血C肽水平升高(P<0.01),其体质量增加(P<0.01),IPGTT结果显示其糖耐量得到改善。结论人外周血内皮细胞与胰岛共移植可明显改善裸鼠移植胰岛的生存。
Objective To investigate the co-transplantation of human peripheral blood endothelial cells and rat islets into the renal capsule of diabetic nude mice to improve the survival and functional development of transplanted islets. Methods Human peripheral blood mononuclear cells (5 × 105) were co-transplanted with freshly isolated rat islet cells into the renal capsule of diabetic nude mice. The levels of fasting blood glucose, body weight and C peptide Level, parallel intraperitoneal glucose tolerance test (IPGTT), to understand the survival of transplanted islets and their functional status. Results Compared with the control group, the blood glucose of the experimental group decreased significantly (P <0.01) and the level of C peptide in the peripheral blood increased (P <0.01), and the body weight increased (P <0.01). The results of IPGTT showed that the glucose tolerance Improved. Conclusion The co-transplantation of human peripheral blood endothelial cells and islets can significantly improve the survival of transplanted islets in nude mice.
