目的探讨参麦注射液(SMI)对阿霉素(ADR)损伤的心肌细胞的保护作用及其机制。方法体外培养的乳鼠心肌细胞,分为正常对照组、ADR损伤模型组和SMI保护组;检测各组心肌细胞培养液中肌酸磷酸激酶(CK)活力、丙二醛(MDA)含量、心肌细胞内超氧化物歧化酶(SOD)活性、一氧化氮(NO)含量及线粒体琥珀酸脱氢酶(SDH)的活力。结果SMI对ADR引起的心肌细胞损伤可减少CK活力、MDA、NO的生成和升高心肌细胞内SOD及线粒体SDH活力。结论SMI对ADR引起的心肌细胞损伤有一定的保护作用,其作用机制与抗脂质过氧化损伤和减少NO生成有关。
Objective To investigate the protective effect of Shenmai injection (SMI) on adriamycin (ADR)-injured cardiomyocytes and its mechanism. Methods Cultured neonatal rat cardiomyocytes were divided into normal control group, ADR injury model group and SMI protection group. Creatine phosphokinase (CK) activity, malondialdehyde (MDA) content, and myocardium were detected in culture fluids of myocardial cells in each group. Superoxide dismutase (SOD) activity, nitric oxide (NO) content, and mitochondrial succinate dehydrogenase (SDH) activity in the cells. RESULTS: SMI caused damage to cardiomyocytes induced by ADR and reduced CK activity, production of MDA and NO, and increased SOD and mitochondrial SDH activity in cardiomyocytes. Conclusion SMI has a protective effect on myocardial cell damage induced by ADR, and its mechanism of action is related to anti-lipid peroxidation injury and reduction of NO production.
